酶类- IMAP 检测技术简介

基于磷酸基团与含三价金属阳离子的纳米微粒(微珠)之间的特异性、高亲和力相互作用,IMAP 是评估激酶、磷酸酶和磷酸二酯酶活性的常用检测技术,且无需抗体。使用荧光标记的底物进行酶活反应。加入 IMAP 结合系统可终止酶反应并引发微珠与磷酸化底物的结合。可以使用 FP 或 TR-FRET 方法读数来检测与酶活性相关的底物与微珠之间的结合情况。

IMAP 改进型结合系统

IMAP 改进型结合系统可让研究人员针对每种底物优化其 FP 和 TR-FRET 的检测条件。该系统由改进型结合缓冲液 A 和改进型结合缓冲液 B 以及改进型结合试剂组成。根据所选底物的酸性特征以及期望的 ATP 浓度和背景参数,可以按不同比例来混合两种缓冲液和试剂。

IMAP 底物

Molecular Devices 可提供多种验证底物和校准品。底物可以与其最初验证底物时所用的酶一起使用,或作为其它酶的潜在底物。
